How to switch from normal mode

  1. Use the 100x / 1.49 oil objective (is the highest numerical aperture lens available)
  2. Switch to the SoRa spinning disc under "pinhole"
  3. Switch to the 2.8x SoRa magnifier
  4. Set the camera to "binning 1x1" (no binning)
  5. Obtain a Z-stack: Make the stack slightly "too big" - having some extra blurry layers above/below the structure helps with deconvolution afterwards.
  6. Perform "3D Deconvolution" (software command) with the standard suggestged settings (mostly change "noisyness" of the picture when needed)
  7. Evaluate result and adjust parameters where necessary (e.g. adjust Z-step size, adjust image "noisyness" setting upon deconvolution)
