How to switch from normal mode
- Use the 100x / 1.49 oil objective (is the highest numerical aperture lens available)
- Switch to the SoRa spinning disc under "pinhole"
- Switch to the 2.8x SoRa magnifier
- Set the camera to "binning 1x1" (no binning)
- Obtain a Z-stack: Make the stack slightly "too big" - having some extra blurry layers above/below the structure helps with deconvolution afterwards.
- Perform "3D Deconvolution" (software command) with the standard suggestged settings (mostly change "noisyness" of the picture when needed)
- Evaluate result and adjust parameters where necessary (e.g. adjust Z-step size, adjust image "noisyness" setting upon deconvolution)
