- 1) Use the 100x / 1.49 oil lens
- 2) Enable the 2.8x SoRa magnifier
- 3) Set camera to "binning 1x1" (no binning)
- 4) Obtain a Z-stack: Include lots of blur above / below the interesting focus layers
- 5) Perform "3D Deconvolution" (software option) with the standard settings.
- 6) Evaluate result and adjust parameters where necessary (e.g. adjust Z-step size, adjust image "noisyness" setting upon deconvolution)
